以化合物(CAS: 624734-16-3)為原料合成3-甲氧基-四氫-2H-吡喃-4-酮的一般步驟如下:將中間體3(3.0 g,17.04 mmol)溶于四氫呋喃/水(60 mL/10 mL)混合溶劑中,加入濃鹽酸(6 mL)。反應(yīng)混合物在室溫下攪拌1小時(shí)。反應(yīng)完成后,將混合物在減壓下濃縮以除去四氫呋喃。隨后,水層用乙醚(6×50 mL)進(jìn)行萃取。合并有機(jī)相,用無(wú)水硫酸鈉干燥,過(guò)濾后減壓蒸發(fā)溶劑,得到中間體24(1.75 g,產(chǎn)率79%),為透明油狀物。產(chǎn)物經(jīng)1H NMR(500 MHz,CDCl3)表征,化學(xué)位移δ為4.23(ddd,J = 1.2, 11.4, 12.4 Hz,1H),4.15-4.09(m,1H),3.82(dd,J = 5.95, 8.7 Hz,1H),3.74(ddd,J = 5.5, 8.5, 13.6 Hz,1H),3.56(dd,J = 8.8, 11.3 Hz,1H),3.50(s,3H),2.61(app dd,J = 5.0, 8.9 Hz,2H)。
