以氫化肉桂酸為原料合成氫化肉桂酰氯的一般步驟:向一個(gè)裝有攪拌棒和氬氣入口隔膜的200 mL燒瓶中加入氫化肉桂酸(5.00 g,33.3 mmol)、N,N-二甲基甲酰胺(DMF,0.1 mL)和二氯甲烷(CH2Cl2,30 mL)。在30分鐘內(nèi),向所得溶液中緩慢加入草酰氯((COCl)2,2 M在CH2Cl2中,20.0 mL,40.0 mmol)。將反應(yīng)混合物在室溫下攪拌過夜。反應(yīng)完成后,通過真空濃縮去除溶劑,得到3-苯基丙酰氯(74,5.9 g,黃色油狀物),收率為100%。通過高效液相色譜(HPLC)分析(流動相比例:水:乙腈:甲醇 = 15:10:75)確認(rèn)產(chǎn)物純度為99%,保留時(shí)間為5.3分鐘。
