E) 奎寧環(huán)-4-基甲胺的合成 將4-氰基奎寧環(huán)(0.30 g,2.2 mmol)溶解于乙醚(5 mL)中,緩慢加入至氫化鋁鋰(LAH,0.167 g,4.41 mmol)的乙醚懸浮液中。反應(yīng)體系在0℃下冷卻后,額外加入乙醚(15 mL)。隨后,將反應(yīng)混合物在23℃下攪拌3小時。反應(yīng)完成后,用硫酸鈉十水合物淬滅反應(yīng),并繼續(xù)攪拌30分鐘。接著,加入乙酸乙酯,30分鐘后,加入硅藻土,通過硅藻土過濾懸浮液。濾液經(jīng)濃縮至干,得到油狀產(chǎn)物奎寧環(huán)-4-基甲胺(0.310 g,定量收率)。該產(chǎn)物無需進(jìn)一步純化,直接用于后續(xù)反應(yīng)。 1H NMR(400 MHz,CDCl3)δ ppm:1.31-1.40(6H,m),2.43(2H,s),2.86-2.94(6H,m)。
