向新制備的乙醇鈉溶液[由乙醇(250mL)與金屬鈉(9.02g,392.55mmol)反應制得]中,依次加入2-氰基-4,4-二乙氧基丁酸乙酯(45g,196.27mmol)和硫脲(14.94g,196.27mmol溶于200mL乙醇)。將反應混合物在攪拌下加熱回流3.5小時。反應完成后,將混合物冷卻至室溫并繼續(xù)攪拌過夜。隨后,用水(100mL)淬滅反應,并通過真空濃縮去除乙醇。將殘余物溶解于水(100mL)中,在溫度低于10℃的條件下,用稀鹽酸水溶液(3N)中和至pH7。過濾收集生成的固體,經(jīng)真空干燥,得到6-氨基-5-(2,2-二乙氧基乙基)-2-巰基嘧啶-4-醇(30.6g,收率60.19%),為淺黃色固體。產物經(jīng)1HNMR、IR、MS、HPLC及元素分析確認。
