以5-溴-1-甲基-3-硝基吡啶-2(1H)-酮為原料合成3-氨基-5-溴-1-甲基吡啶-2(1H)-酮的一般步驟:在氮?dú)獗Wo(hù)下,向裝有機(jī)械攪拌器和回流冷凝器的1升三頸圓底燒瓶中加入5-溴-1-甲基-3-硝基吡啶-2(1H)-酮(21.7g,93.3mmol)、乙醇(305mL)、鐵粉(-325目,52.1g,933mmol)和2N鹽酸(50mL,100mmol)。將反應(yīng)混合物在60℃下加熱攪拌2小時(shí)。反應(yīng)完成后,冷卻至室溫,加入碳酸鉀調(diào)節(jié)pH至8(通過(guò)pH試紙測(cè)定)。過(guò)濾所得懸浮液,濾餅用乙醇(4×100mL)洗滌。合并濾液,減壓濃縮,得到棕色固體。該固體通過(guò)硅膠柱色譜純化,得到目標(biāo)產(chǎn)物3-氨基-5-溴-1-甲基吡啶-2(1H)-酮(14.5g,收率77%),為灰白色粉末,熔點(diǎn)為104-105℃。1H NMR(500MHz,DMSO-d6)δ7.15(d,1H,J=2.5Hz),6.46(d,1H,J=2.5Hz),5.45(bs,2H),3.40(s,3H);MS(ESI+)m/z 203(M+H)。
