在密封的玻璃管中,將甲醛(1g,33.54mmol)和1-甲基-5-硝基-1H-咪唑(4a)(1g,7.8mmol)溶解于DMSO(5mL)中。將反應(yīng)管置于140℃油浴中加熱48小時(shí)。反應(yīng)完成后,將反應(yīng)液倒入水(5mL)中,并用乙酸乙酯(3×5mL)進(jìn)行萃取。合并有機(jī)相,用無(wú)水硫酸鎂干燥,隨后在減壓下濃縮除去溶劑,得到粗產(chǎn)物。粗產(chǎn)物通過(guò)THF/庚烷(1:1)混合溶劑進(jìn)行重結(jié)晶純化,得到白色粉末狀的目標(biāo)產(chǎn)物1-甲基-5-硝基-2-羥甲基咪唑(5a)(0.76g,收率62%)。產(chǎn)物的熔點(diǎn)為117°C。
