頭孢氨芐EP雜質(zhì)C性質(zhì)、用途與生產(chǎn)工藝
頭孢氨芐為半合成的第一代口服頭孢菌素,是對(duì)革蘭陽(yáng)性、陰性細(xì)菌都有殺菌作用的廣譜抗生素。臨床適用于泌尿道、呼吸道、皮膚和軟組織、生殖器官(包括前列腺)等部位的感染,也常用于中耳炎。頭孢氨芐EP雜質(zhì)C是頭孢氨芐重要的雜質(zhì)。
將N-BOC-鄰苯基甘氨酸(753mg,3mmol)溶解在四氫呋喃(THF)(15ml),Et3N0.42ml(3mmol)和3滴N-甲基嗎啉的混合物中,在-15℃下攪拌10分鐘。逐滴加入在2mlTHF中的氯甲酸異丁酯(0.41ml,3mmol)。在-15°C攪拌20分鐘后,將1.1g(3mmol)頭孢氨芐溶解在10ml含0.42in1(3mmol)Et,N的THF-H2O(1:l)中。加入。將混合物在0℃下攪拌3小時(shí)。通過蒸發(fā)除去THF。加入H2O(10ml),并用2NHCl將pH調(diào)節(jié)至2。然后將溶液用EtOAc(3×20ml)萃取。合并的組織。將水層以H2O(20毫升)洗滌,以無(wú)水MgSO4干燥,過濾并蒸發(fā)。將殘余物溶解在約2ml的氟乙酸中,并在室溫下保存。持續(xù)16小時(shí)。蒸發(fā)溶液,將溶于水的殘余物置于陰離子交換柱(Bio-RadAG-3,200-400目,乙酸鹽,13×2cm)上,并用水洗脫以除去三氟乙酸。蒸發(fā)洗脫液,殘余物從丙酮-水中結(jié)晶并真空干燥。頭孢氨芐EP雜質(zhì)C產(chǎn)率900mg(63%)。
