一般步驟:將7-甲基-4-苯基-3,4-二氫香豆素(9.52 g,40.0 mmol)的無水四氫呋喃(50 mL)溶液緩慢滴加至冷卻至0℃的氫化鋁鋰(3.04 g,80 mmol)在無水四氫呋喃(100 mL)中的懸浮液中。反應(yīng)混合物在室溫及惰性氣氛下攪拌1小時(shí)。隨后,謹(jǐn)慎加入50 mL四氫呋喃與水的1:1混合液,并用1 mol/L鹽酸溶液(150 mL)酸化以淬滅反應(yīng)。用乙酸乙酯萃取反應(yīng)產(chǎn)物,合并有機(jī)相,依次用水和鹽水洗滌,無水硫酸鎂干燥。減壓除去溶劑后,得到2-(3-羥基-1-苯基丙基)-4-甲基苯酚(式II化合物),收率97%(9.68 g)。產(chǎn)物通過二異丙醚重結(jié)晶,獲得白色粉末。熔點(diǎn):112-115℃。1H NMR (300 MHz, DMSO-d6): δ [ppm] = 2.08-2.17 (5H, m, CH3, CH2), 3.29-3.31 (2H, m, CH2), 4.38, 4.42 (2H, 2 xt, CH, OH), 6.63 (1H, d, J = 8.0 Hz, 1H-Ar), 6.76 (1H, dd, J = 8.3, 1.7 Hz, 1H-Ar), 7.00 (1H, d, J = 1.7 Hz, 1H-Ar), 7.08-7.13 (2H, m, 1H-Ph), 7.20-7.28 (3H, m, 4H-Ph), 9.04 (1H, s, OH). 13C NMR (300 MHz, DMSO-d6): δ [ppm] 20.5, 37.6, 39.1, 59.3, 115.0, 125.6, 127.0, 127.3, 127.9, 128.0, 130.9, 145.3, 152.4. ESI-MS: 243 [M + H]+.
