| In vitro | Antitumor agent-53 (compound 6f) exhibited dose-dependent anti-proliferative activity across various cell lines (HGC-27, HT-29, HepG-2, A549, MCF7, GES-1) with IC50 values ranging from 0.37 to >18 μM after 72 hours of treatment. Particularly, it inhibited cell proliferation in HGC-27 and HT-29 cells at concentrations of 0.15, 0.3, and 0.6 μM and demonstrated inhibitory effects on Topo I at 200 μM. The compound induced G2/M phase cell cycle arrest in HGC-27 and HT-29 cells at 0.1, 0.3, and 0.9 μM after 24 hours, promoted apoptosis, and inhibited cell migration and invasion in these cell lines in a concentration-dependent manner, with effects observed from 0.1 to 2.7 μM over 24 hours. It also suppressed the PI3K/AKT pathway, further inducing apoptosis in HGC-27 cells at 0.1, 0.3, and 0.9 μM, demonstrating its potential as a multifaceted antitumor compound. |
