描述	L-DOPA belongs to the category of dopamine precursors, serving as an orally active neurotransmitter metabolic precursor capable of crossing the blood-brain barrier and undergoing conversion to dopamine within the brain. The compound exhibits anti-hyperalgesic properties and finds application in Parkinson's disease research as well as in the induction of disease models.
激酶實驗	Briefly, transfected HEK-293 cells, incubated in charcoal-treated Dulbecco's modified Eagle's medium for 24 h, are washed once with Hanks' solution and resuspended in a buffer containing 100 mM NaCl, 1 mM MgCl2, 1 mM EDTA, 1 mM EGTA, 250 mMsucrose, 20 mM Tris-HCl, pH 7.4. Cells are lysed by freezing in liquid nitrogen. Dehydrogenase activity is measured in a final volume of 20 μL containing the appropriate concentration of bile acid, 30 nCi of [3H]cortisol, and unlabeled cortisol to a final concentrations of 50 nM. The reaction is started by mixing cell lysate with the reaction mixture. Alternatively, endoplasmic reticulum microsomes are prepared from transfected HEK-293 cells and incubated with reaction mixture containing various concentrations of cortisol and CDCA. Incubation proceeded for 20 min, and the conversion of cortisol to cortisone is determined by thin layer chromatography (TLC). Because of the inaccuracy of the TLC method at low conversion rates and the end-product inhibition of 11βHSD2 at conversion rates higher than 60-70%, only conversion rates between 10 and 60% are considered for calculation. The inhibitory constant IC50 is evaluated using the curve-fitting program. Results are expressed as means±S.E. and consist of at least four independent measurements.
體外活性	方法：采用C6膠質(zhì)肉瘤細胞，分別通過體外存活實驗（孵育硼濃度20 ppm的1?B，持續(xù)6 h）、細胞毒性實驗（連續(xù)暴露3 d檢測IC50）及硼攝取實驗（1.5 mM硼試劑培養(yǎng)24 h）評估碳硼烷基L-DOPA的抗腫瘤活性。結(jié)果：碳硼烷基L-DOPA在熱中子照射75 min后最低存活分數(shù)為0.03，IC50為8.93×10?? M，細胞硼攝取量為1.53 μg/10?細胞，顯示出優(yōu)于BSH的體外殺傷效果。[1]
體內(nèi)活性	方法：采用6-OHDA損毀單側(cè)黑質(zhì)紋狀體通路的大鼠模型，給予左旋多巴（50 mg/kg，腹腔注射，聯(lián)合芐絲肼12.5 mg/kg，每日兩次，連續(xù)5-15天），通過原位雜交和放射自顯影檢測D3受體表達，并觀察旋轉(zhuǎn)行為；同時采用MPTP誘導的帕金森猴模型，給予左旋多巴觀察異常運動。結(jié)果：左旋多巴重復給藥誘導去神經(jīng)支配的尾殼核出現(xiàn)D3受體異位表達，該表達與旋轉(zhuǎn)行為增強（行為敏化）呈平行時間過程，同時伴有前強啡肽原mRNA上調(diào)和前速激肽原mRNA下調(diào)；在猴模型中引起多種異常運動。[2] 方法：給完整大鼠注射L-DOPA（50 mg/kg）檢測基底節(jié)區(qū)內(nèi)酰胺濃度；另在損傷大鼠中用L-DOPA誘導口-舌自發(fā)運動，并用大麻素激動劑R(+)-WIN55,212-2（1 mg/kg）處理。結(jié)果：L-DOPA增加基底節(jié)區(qū)內(nèi)酰胺濃度；誘導日益嚴重的口-舌自發(fā)運動，大麻素激動劑可減緩該運動。[3] 方法：采用6-OHDA誘導的單側(cè)重度損傷大鼠模型，通過口服給予左旋多巴（25 mg/kg，聯(lián)合卡比多巴，每日兩次）連續(xù)6個月，使用125I-磺必利放射自顯影檢測紋狀體D2受體結(jié)合水平。結(jié)果：慢性左旋多巴給藥逆轉(zhuǎn)了重度損傷大鼠尾殼核（尤其是背外側(cè)和腹外側(cè)區(qū)域）的D2受體上調(diào)，證實其在基底節(jié)達到生物活性濃度。[4]
