1.1 1. Synthesis of 3-((N-(methoxycarbonyl)methyl)sulfonyl)-2-thiophenecarboxylic acid methyl ester
The four-mouth flask equipped with a stirrer was added methyl 3-chlorosulfonyl-2-thiophenecarboxylate 8 g and water 20 ml. At room temperature, add dropwise glycine methyl ester hydrochloride 4.5 g aqueous solution (10 ml). At the same time, add dropwise 10% acid binding agent aqueous solution 30 ml, control pH to 7-8. After the completion of the addition, reaction at room temperature for 6 hours, filtering, the filter cake after the water washing, drying after recrystallization with methanol, getting white solid TNXK-1 to 9 g, yield 92.3%.
