add 1.1 g (6.0 mmol) to the three-necked flask4-aminophthalic acid prepared in step (1), and then added 1.39 g in turn(12.6 mmol) resorcinol, 10 mL methanesulfonic acid, under argon,The reaction was carried out at 97 °C for 3 h, cooled to room temperature, and the resulting reaction solution was poured into 30 mL of ice water.The pH of the system was adjusted to 6 with 5 mol/L NaOH solution, and a reddish-brown solid was precipitated.After refrigerating at 5°C for 2 hours, no solid was precipitated, suction filtered, and the filter cake was washed with water until neutral.Vacuum dried to constant weight at 50°C to obtain 2.03 g of a tan solid,It is 5-aminofluorescein with a yield of 94.5%.
