I.2 Step 2: 5-iodo-2-(methylthio')pyrimidin-4(3H)-one (3)
Step 2: 5-iodo-2-(methylthio')pyrimidin-4(3H)-one (3) To a solution of 2-(methylthio)pyrimidin-4(3H)-one (2) (4.74 g, 33.3 mmol) in chloroform (45 mL), NIS (8.25 g, 36.7 mmol) was added. The solution was heated to 70 °C for 2 hours. The mixture was concentrated under reduced pressure. EtOAc was added and the organics were washed with water, filtered and dried to give the title compound (8.8 g, 97%) as a white solid. NMR (CD3OD) δ 8.30 (s, 1H), 2.45 (s, 3H); LCMS (Condition B): 1.43 min, 269 (M+H).
