53.4 Synthesis of ethyl 3-isocyanatopropanoate (5)
β- Alanine ethyl ester HCl (263 mg, 1.71 mmol) in CH2C12 (8 mL) and saturated NaHCO3 solution (8 mL) was degassed and the vial was cooled in ice water bath. Triphosgene (508 mg, 1.71 mmol) was added in one portion under inert atmosphere at 0 °C. The reaction stirred at 0 °C to RT over 3 hr. The reaction was diluted with water (15 mL), the layers separated and the aqueous layer was extracted with CH2Cl2. The combined organic extracts were washed with brine, dried over Na2S04, filtered, and concentrated under reduced pressure to obtain crude 5 (216 mg) as a white crystalline solid. This crude material was directly used for next reaction without purification. 1H NMR (300 MHz, CDCl3): δ 4.20 (q, 2H), 3.59 (t, 2H), 2.60 (t, 2H), 1.28 (t, 3H).
