Example 1 Preparation of 1-cyclopropyl-6-fluoro-7-((4aS,7aS)-hexahydro-1H-pyrrolo[3,4-b]pyridin-6(2H)-yl)-8-methoxy-4-oxo-1,4-dihydroquinoline-3-carboxylic acid. In dimethyl sulfoxide (600 mL), 50 g of 1-cyclopropyl-6,7-difluoro-8-methoxy-4-oxo-1,4-dihydro-3-quinolinecarboxylic acid and 25.70 mL of (4aS,7aS)-octahydro-1H-pyrrolo[3,4-b]pyridine were added and the reaction was stirred for 6-8 hr at 65-70 °C. The reaction process was monitored by HPLC. Upon completion of the reaction, the reaction mixture was cooled to 25-30 °C, 100 mL of water was added and stirring was continued for 2 hours. The solid product was collected by diafiltration, washed with 100 mL of water and subsequently dried under vacuum at 75 °C to give 61.0 g of the target compound. Yield: 89.7%; Purity: 98% (HPLC); Chiral impurity ((R,R)-isomer) content: 3-5%.
