4 Production Example 4: Synthesis of Intermediate D-12
A solution of 2 -bromopropane (300 g, 2.44 mol) , 2,2- diethoxyethanamine (341 g, 2.56 mol) and potassium carbonate (674 g, 4.88 mol) in MeCN (3.0 L) was stirred at 80°C for 16 hrs. The solution was filtered and the filtrate was concentrated. The crude product was dissolved in ethyl acetate (2.0 L) , washed with water (1.0 L) and brine (1.0 L) ; dried over anhydrous sodium sulfate, and then filtered and evaporated to give the desired product of D-12 (300 g, 1.71 mol, 70% yield) as yellow oil, which was used for next step without purification.XH NMR (400 MHz, CDCI3) data of D-12 is shown in Fig. 5.
