在恒定攪拌下,將2-羥基-5-硝基吡啶(200 g,1.42 mol)分批加入800 mL濃鹽酸中,隨后加熱至50℃。緩慢滴加氯酸鈉水溶液(80.0 g,0.75 mol溶于適量水)。維持反應(yīng)溫度在50℃繼續(xù)攪拌1小時(shí),反應(yīng)完成后冷卻至0℃。過濾收集析出的固體產(chǎn)物,用冷水充分洗滌,干燥后得到2-羥基-3-氯-5-硝基吡啶。產(chǎn)量:240 g,1.37 mol(產(chǎn)率96.0%)。熔點(diǎn):195-197℃。1H NMR(300 MHz,DMSO-d6):δ 8.65(d,1H,J = 2.5 Hz),8.36(d,1H,J = 2.5 Hz)。13C NMR(300 MHz,DMSO-d6):δ 153.40,144.29,143.59,135.08,130.44。質(zhì)譜(ES):m/z 175([M + H]+)。
