步驟B. 在0℃下,將4-氟-3-硝基苯胺(5.0g,32.0mmol)和三乙胺(TEA,6.7mL,48.0mmol)溶解于50mL二氯甲烷(DCM)中。隨后,緩慢滴加乙酰氯(2.75mL,38.4mmol),滴加完畢后將反應(yīng)混合物升溫至室溫并攪拌過夜。反應(yīng)完成后,依次用5%硫酸氫鉀(KHSO4)水溶液、飽和碳酸氫鈉(NaHCO3)水溶液和鹽水洗滌有機(jī)層,然后用無水硫酸鎂(MgSO4)干燥。通過濃縮有機(jī)相并從DCM中結(jié)晶,得到N-(4-氟-3-硝基苯基)乙酰胺5.3g,產(chǎn)率84%。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR(400MHz,氯仿-D)確認(rèn):δ2.04(s,3H),7.51(dd,J = 11.23,9.08Hz,1H),7.80(ddd,J = 9.08,4.00,2.93Hz,1H),8.47(dd,J = 7.03,2.73Hz,1H),10.38(s,1H)。
