實施例1：2-[6-(1-溴-乙基)-5-氟嘧啶-4-基-硫烷基]苯甲酸甲酯的制備（鄰位取代-CO2CH3的式2化合物的制備）；步驟1）2-巰基苯甲酸甲酯的制備：將40g 2-巰基苯甲酸溶于600mL甲醇中，冷卻至5°C。在10分鐘內緩慢滴加42mL濃硫酸。反應混合物在回流條件下反應14小時后，進行減壓濃縮。濃縮物用400mL二氯甲烷和400mL純水稀釋，隨后分離有機層。有機層依次用200mL飽和碳酸氫鈉溶液和400mL飽和食鹽水洗滌，然后用無水硫酸鈉干燥。干燥后，有機層經濃縮得到粗產物，進一步通過分餾純化（118°C/7mmHg）。最終獲得42g（產率：96%）硫代水楊酸甲酯，為透明油狀物。1H-NMR（200MHz，CDCl3）δ（ppm）：8.01（d，1H），7.30（d，2H），7.14（m，1H），4.68（s，1H），3.91（s，3H）。
