以6-喹啉羧酸甲酯為原料合成6-羥甲基喹啉的一般步驟:在-78℃的丙酮-干冰浴中冷卻0.2M的6-喹啉羧酸甲酯(0.300g,1.60mmol)的THF(8mL)溶液和0.6M的二異丁基氫化鋁(DIBAL)(0.860mL,4.81mmol)的THF溶液(8mL)。通過套管將DIBAL溶液轉(zhuǎn)移至甲酯溶液中,所得溶液在0℃下攪拌1小時。反應(yīng)通過在0℃下加入甲醇(8mL)和乙酸(1.37mL,24.0mmol)淬滅。攪拌5分鐘后,加入飽和酒石酸鈉溶液(16mL)。繼續(xù)攪拌20分鐘,隨后用EtOAc(50mL)和水(10mL)稀釋反應(yīng)混合物。水層用EtOAc(3×50mL)萃取。合并有機(jī)相,用15mL水洗滌一次。有機(jī)層用Na2SO4干燥,過濾后減壓濃縮。通過柱色譜(50-90%EtOAc/己烷)純化,得到0.236g(93%)的6-羥甲基喹啉(S3a),為淡黃色油狀物。1H NMR(500MHz,CDCl3):δ4.89(s,2H),7.35(dd,1H,J=4.0,8.0),7.65(dd,1H,J=1.5,9.0),7.77(s,1H),8.01(d,1H,J=9.0),8.08(d,1H,J=4.0),8.80(dd,1H,J=1.5,4.0)。13C-NMR(125MHz,CDCl3):δ64.8,121.5,125.0,128.3,129.0,129.4,136.4,139.9,147.7,150.2。HRMS-FAB(m/z):[MH]+ C10H10NO的計算值,160.0762;實(shí)測值:160.0766。
